YSA peptide-modified adenovirus is a genetically engineered adenoviral vector with the YSA peptide (Tyr-Ser-Ala) inserted into its fiber HI-loop. The YSA peptide specifically binds to the EphrinA2 (EphA2) receptor, which is frequently overexpressed on various tumor cells, notably pancreatic cancers. This modification redirects the tropism of the adenoviral vector from its native coxsackie and adenovirus receptor (CAR) tropism to EphA2, thereby enhancing transduction efficiency in EphA2-positive tumor cells and reducing dependency on CAR expression. The technology is used primarily to improve gene delivery or oncolytic viral therapies targeting tumors that overexpress EphA2, especially pancreatic and potentially other solid malignancies. The modification may be used for cancer gene therapy or oncolytic virotherapy applications[2][5][10].
