YT-DP is a glyco-conjugation-based dual-payload antibody-drug conjugate (ADC) platform developed by Glyco-therapy Biotechnology. The platform utilizes the proprietary YTConju glyco-conjugation system to incorporate two distinct mechanism-of-action (MOA) payloads into a single antibody molecule through a one-step conjugation strategy. Specifically, it integrates a cleavable-linker-topoisomerase I inhibitor payload (YT-CLTp) with a microtubule inhibitor (MTI). This dual-payload approach is designed to enhance antitumor efficacy, achieve synergistic effects, and overcome tumor heterogeneity and therapeutic resistance. The platform allows for flexible drug-to-antibody ratio (DAR) configurations, such as DAR 2+2 or DAR 2+4, and has demonstrated superior potency compared to standard ADCs like DS-8201 in preclinical models. Preclinical studies using trastuzumab-based dual-payload ADCs (Tras-DPs) have shown high homogeneity, excellent stability, and potent bystander killing effects.
