Drug intelligence / Profile preview
YTH24.5-SG3249 is a preclinical-stage antibody-drug conjugate (ADC) consisting of the rat IgG2b anti-human CD45 monoclonal antibody clone YTH24.5 conjugated to the cytotoxic pyrrolobenzodiazepine (PBD) dimer payload tesirine (SG3249) via a cathepsin B-cleavable valine-alanine linker. Developed by ADC Therapeutics in collaboration with UCL Great Ormond Street Institute of Child Health, UCL Cancer Institute, and Great Ormond Street Hospital, the ADC targets CD45 (receptor-type tyrosine-protein phosphatase C), which is expressed on almost all cells of the hematopoietic lineage, including hematopoietic stem cells (HSCs) and hematological malignancies. YTH24.5-SG3249 is designed as a targeted conditioning agent for allogeneic stem cell transplantation (SCT) and gene therapy, aiming to ablate host HSCs with minimal non-hematopoietic toxicity, and as a cytoreductive bridge to SCT in patients with refractory leukemia.
