Overview

YTH54.12 is an unconjugated rat monoclonal antibody targeting the human CD45 antigen (receptor-type tyrosine-protein phosphatase C), which is expressed on all hematopoietic cells except mature red blood cells and platelets. Developed by Baylor College of Medicine, it is primarily investigated for use in conditioning regimens prior to allogeneic hematopoietic stem cell transplantation in patients with advanced leukemia and myelodysplastic syndromes (MDS). By binding to CD45, YTH54.12 facilitates the depletion of host lymphocytes and hematopoietic progenitors, thereby reducing the risk of graft rejection and promoting donor cell engraftment. In clinical trials, it is frequently evaluated in combination with another anti-CD45 antibody, YTH25.4, as well as chemotherapy and total body irradiation.
