Overview

Yttrium-90-labeled anti-tenascin monoclonal antibody (specifically the 81C6 clone) is an investigational radiopharmaceutical developed by Duke University Medical Center for the treatment of malignant gliomas, including glioblastoma and anaplastic astrocytoma. This radioimmunotherapeutic agent consists of a monoclonal antibody that specifically targets Tenascin-C, an extracellular matrix glycoprotein highly overexpressed in the stroma of various tumors but minimally expressed in normal adult brain tissue. The antibody is conjugated to yttrium-90, a high-energy beta-emitting radioisotope. For brain tumor therapy, the drug is typically administered via intracavitary injection directly into the surgical resection cavity following tumor removal. This local administration bypasses the blood-brain barrier and allows for the targeted delivery of cytotoxic radiation to residual cancer cells at the tumor margin while minimizing systemic exposure and damage to surrounding healthy brain tissue.
