Overview

yttrium-90-DOTA-M5A is a radioimmunoconjugate composed of the humanized monoclonal antibody M5A, which targets carcinoembryonic antigen (CEA), conjugated to the radioactive isotope yttrium-90 via a DOTA chelator. The drug is designed for targeted radioimmunotherapy of CEA-expressing tumors, such as colorectal cancer and medullary thyroid cancer. By binding to CEA on tumor cells, it delivers cytotoxic radiation directly to malignant tissue while sparing normal cells. The humanized nature of the M5A antibody reduces immunogenicity compared with murine or chimeric antibodies, potentially allowing for repeated dosing. Developed initially by the National Cancer Institute, it has been evaluated in phase I/II clinical trials primarily for advanced metastatic cancers expressing CEA[1][2][4][6].
