yttrium-90-DOTA-rat immunoglobulin G is an experimental radiolabeled antibody construct used primarily as a non-specific isotype control in preclinical radioimmunotherapy (RIT) research. It consists of polyclonal rat immunoglobulin G (IgG) conjugated to the chelator DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) and labeled with the high-energy beta-emitting radioisotope yttrium-90 (90Y). In studies evaluating targeted therapies for hematologic malignancies like acute myeloid leukemia (AML), this construct is used to establish a baseline for non-specific radiation-induced toxicity and biodistribution. Because it lacks a specific biological target, its effects are mediated through the systemic emission of beta particles, which can cause dose-dependent myelosuppression and other toxicities, serving as a critical comparator for assessing the therapeutic index of targeted agents such as anti-CD45 antibodies.
