Overview

Yttrium-90-labeled anti-CEA antibody is a radiolabeled monoclonal antibody therapy designed for targeted radioimmunotherapy of cancers expressing carcinoembryonic antigen (CEA). The drug consists of a humanized or chimeric monoclonal antibody that specifically binds to CEA on tumor cells and is conjugated to the radioactive isotope yttrium-90 via a chelator such as DOTA or DTPA. Upon binding to CEA-expressing cells, the beta-emitting yttrium-90 delivers localized radiation intended to destroy malignant cells while sparing surrounding healthy tissue. Several variants exist based on the specific mAb used (e.g., M5A, hMN-14/labetuzumab, cT84.66). This approach has been investigated primarily in advanced solid tumors such as colorectal cancer and medullary thyroid cancer[1][2][3][6]. The mechanism combines targeted immunotherapy with radiopharmaceutical cytotoxicity.
