Overview

Yttrium-90-mCC49 is a radiolabeled monoclonal antibody therapy that targets tumor-associated glycoprotein 72 (TAG-72), an antigen commonly expressed on the surface of various adenocarcinomas, including gastrointestinal and non-small cell lung cancers. The drug consists of the murine monoclonal antibody CC49 conjugated to the radioactive isotope yttrium-90, enabling targeted delivery of beta radiation to TAG-72–positive tumor cells. Its mechanism involves binding to TAG-72 on cancer cells, allowing localized radiation-induced cytotoxicity while sparing most normal tissues. Clinical studies have evaluated its safety and maximum tolerated dose in patients with advanced cancers, often in combination with agents such as interferon alpha or paclitaxel for radiosensitization. Due to immunogenicity concerns with murine antibodies, humanized versions are under investigation[1][5].
