YY-20394 (linperlisib) is a small molecule, highly selective inhibitor of phosphatidylinositol 3-kinase delta (PI3Kδ), developed as an oral antineoplastic agent targeting B-cell malignancies. Gemcitabine is a nucleoside analog functioning as an antimetabolite chemotherapeutic, and oxaliplatin is a platinum-based chemotherapeutic that causes DNA crosslinking and apoptosis. The triplet regimen of YY-20394 + gemcitabine + oxaliplatin is under investigation as an anti-cancer combination regimen for hematologic and possibly solid tumors, particularly in relapsed/refractory lymphoma and other cancers. YY-20394 primarily inhibits PI3Kδ, impacting B-cell proliferation and survival[3][5]. Gemcitabine inhibits DNA synthesis, and oxaliplatin induces cytotoxic DNA damage[2][4][6].
