YZ-0231 is a small molecule inhibitor of the NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) signaling pathway, currently under development by Guangdong Yinzhu Pharmaceutical Technology. The drug is primarily being investigated for the treatment of EGFR-positive non-small cell lung cancer (NSCLC), specifically in combination with the third-generation EGFR tyrosine kinase inhibitor (TKI) osimertinib. The rationale for this combination is to target NF-κB activation, which has been identified as a key mechanism of acquired resistance to EGFR-TKIs. By inhibiting NF-κB, YZ-0231 aims to restore sensitivity to osimertinib and improve clinical outcomes in patients with advanced NSCLC who have progressed on prior TKI therapy. It is currently in Phase 2 clinical development.
