zabadinostat + PD-1 inhibitor

Zabadinostat + PD-1 inhibitor is a combination therapy being developed by IngeneOx Therapeutics for the treatment of advanced solid tumors, specifically microsatellite stable (MSS) colorectal cancer and hepatocellular carcinoma (HCC). Zabadinostat (also known as CG-745) is a small molecule inhibitor of histone deacetylases (HDAC) that modulates the tumor microenvironment by increasing the expression of tumor antigens and MHC molecules while reducing the activity of immunosuppressive cells like MDSCs and Tregs. By combining zabadinostat with a PD-1 checkpoint inhibitor, the therapy aims to overcome primary or acquired resistance to immunotherapy, enhancing T-cell infiltration and anti-tumor immune responses. The combination is currently in Phase 2 clinical development under the program names HACCA and CAROSELL.
