Zabinostat + anti-PD1 is a combination therapy comprising zabinostat (CXD101), a selective class I histone deacetylase (HDAC) inhibitor, and a programmed cell death protein 1 (PD-1) blocking monoclonal antibody. Developed by Celleron Therapeutics (and its affiliate Tcellaron Therapeutics), the combination is designed to overcome immunotherapy resistance in "cold" tumors. Zabinostat acts as an epigenetic modulator that enhances the expression of immune-related genes, increases MHC class I and II molecules on tumor cells, and promotes the infiltration and activity of T lymphocytes and dendritic cells. This priming effect is intended to sensitize microsatellite-stable (MSS) colorectal cancer (CRC) and other refractory malignancies to PD-1 checkpoint inhibition. The combination has been investigated in Phase 2 and Phase 3 clinical trials, particularly for MSS CRC and B-cell lymphomas.
