Overview

Zafirlukast (ICI 204,219) is an orally active small molecule that acts as a potent and selective antagonist of the leukotriene D4 (LTD4) receptor. It inhibits the action of cysteinyl leukotrienes at the CysLT1 receptor on target cells such as airway smooth muscle and eosinophils. By blocking this pathway, zafirlukast reduces bronchoconstriction and inflammation associated with asthma and allergic rhinitis. It was developed primarily for the treatment of asthma but has also been studied for seasonal allergic rhinitis[2][3][4][5]. The drug improves both objective (e.g., FEV1) and subjective measures of asthma severity in clinical trials[2][7].
