Overview

ZAG-101 is a bifunctional antibody drug being developed by Zag Bio for the treatment of Type 1 diabetes. The therapeutic approach involves delivering pancreatic beta cell antigens directly to the thymus, the organ responsible for educating T cells. By presenting these antigens within the thymic environment, ZAG-101 aims to engage the body's natural machinery for central immune tolerance, teaching the immune system to recognize beta cells as self rather than foreign targets. This strategy is designed to halt the autoimmune progression of Type 1 diabetes and preserve remaining beta cell function. Zag Bio, based in Cambridge, Massachusetts, is supported by significant Series A funding from investors including Polaris Partners, the T1D Foundation, Sanofi, AbbVie, and Regeneron, with plans to enter clinical trials by late 2026.
