Overview

Zalifrelimab is a fully human monoclonal antibody of the IgG1 subtype that targets cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), an immune checkpoint protein expressed on T-cells. By binding to and blocking CTLA-4, zalifrelimab inhibits its interaction with ligands CD80 and CD86, thereby reducing CTLA-4-mediated inhibition of T-cell activation. This mechanism enhances the immune system’s ability to recognize and destroy cancer cells by increasing the number of activated cytotoxic T-cells in the tumor microenvironment. Zalifrelimab is being developed as an immunotherapy for various solid tumors, including cervical cancer, non-small cell lung cancer, soft tissue sarcoma, pancreatic cancer, prostate cancer, bladder cancer, and others. It is currently under investigation in multiple Phase I/II clinical trials for these indications[1][2][3][4][5][6][7].
