Overview

Zalutumumab is a fully human IgG1 monoclonal antibody that selectively targets the epidermal growth factor receptor (EGFR). By binding to the extracellular domain III of EGFR, it locks the receptor in an inactive conformation, preventing dimerization and subsequent intracellular signaling. This results in inhibition of tumor cell proliferation and survival. Additionally, zalutumumab mediates antibody-dependent cellular cytotoxicity (ADCC), contributing to its antitumor effects. It was developed primarily for squamous cell carcinoma of the head and neck (SCCHN) but has also been investigated for non-small cell lung cancer and colorectal cancer. Zalutumumab was developed by Genmab but development has been discontinued after clinical trials failed to show significant benefit over standard therapies[1][2][3][6].
