**Zalutumumab + cisplatin + docetaxel** is a combination regimen used experimentally in the treatment of squamous cell carcinoma of the head and neck (SCCHN) and other cancers. Zalutumumab is a fully human IgG1 monoclonal antibody directed against epidermal growth factor receptor (EGFR), blocking ligand binding and promoting antibody-dependent cellular cytotoxicity (ADCC)[2][4][8]. Cisplatin is a platinum-based chemotherapy agent that causes DNA crosslinking, leading to apoptosis; docetaxel is a taxane that disrupts microtubule function, inhibiting cell division. This combination aims to synergistically inhibit tumor growth through EGFR blockade, DNA damage, and mitotic inhibition. Trials have explored this regimen in advanced and recurrent/metastatic SCCHN, particularly when standard therapies have failed or are not tolerated[7][2][4][8]. Zalutumumab clinical development was suspended due to lack of clear additional benefit and toxicity, but the principle of combined EGFR inhibition with platinum/taxane chemotherapy has been investigated.
