Zamaglutenase (TAK-062, Kuma062) is an investigational, orally administered, computationally engineered recombinant protease being developed by Takeda for the treatment of celiac disease. Originally designed by PvP Biologics, the drug is a glutenase derived from the bacterial enzyme kumamolisin-As from *Alicyclobacillus sendaiensis*. It is specifically engineered to remain stable and active in the acidic environment of the stomach, where it targets and degrades immunogenic gluten peptides by cleaving proline-glutamine (P-Q) dipeptide motifs. By enzymatically neutralizing more than 99% of ingested gluten before it reaches the duodenum, zamaglutenase aims to prevent the T-cell mediated immune response and subsequent intestinal mucosal damage in celiac patients who are exposed to gluten despite maintaining a gluten-free diet.
