Overview

Zamristobart (SAR-444881) is a humanized IgG4κ monoclonal antibody designed to target the Leukocyte Immunoglobulin-Like Receptor Subfamily B Member 1 (LILRB1), also known as ILT2 or CD85j. LILRB1 is an inhibitory immune checkpoint receptor expressed on various immune cells, including NK cells, T cells, B cells, dendritic cells, and macrophages. It suppresses immune activity by binding to classical and non-classical MHC class I molecules, such as HLA-G and beta-2 microglobulin (B2M), a mechanism that cancer cells exploit to evade immune destruction. By blocking the LILRB1–MHC class I interaction, zamristobart aims to restore immune activation, enhance phagocytosis, and exert antineoplastic effects. Developed by Sanofi, the drug is currently being investigated in clinical trials for the treatment of malignancies such as chronic lymphocytic leukemia (CLL) and multiple myeloma (MM).
