Zan-81 is an antibody-drug conjugate (ADC) being developed by Corellia AI for the treatment of HER2-positive relapsed or refractory breast cancer, specifically targeting patients who have progressed after treatment with trastuzumab deruxtecan (Enhertu). The drug utilizes Corellia AI's proprietary Coromip technology to conjugate a HER2-targeting monoclonal antibody with Omipalisib (also known as compound '81'), a potent dual inhibitor of PI3K and mTOR. By delivering the PI3K/mTOR inhibitor directly to HER2-expressing cancer cells, Zan-81 aims to overcome resistance mechanisms associated with the PI3K/AKT/mTOR pathway while minimizing systemic toxicity. It is currently in the discovery and preclinical stages of development.
