This is a combination therapy consisting of **zanidatamab**, **docetaxel**, and **carboplatin**. Zanidatamab is a bispecific (biparatopic) monoclonal antibody that targets two distinct extracellular domains on the HER2 receptor, inducing distinct HER2 clustering, receptor internalization, downregulation, and inhibition of downstream signaling pathways leading to cancer cell growth inhibition, antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC)[1][3][5][7]. Docetaxel is a taxane-class small molecule chemotherapeutic that stabilizes microtubules and inhibits their disassembly, preventing cell division and inducing apoptosis. Carboplatin is a platinum-based small molecule chemotherapeutic that induces DNA crosslinking and apoptosis by interfering with DNA replication and transcription. This combination is under active clinical investigation for HER2-positive cancers, including gastroesophageal adenocarcinoma and other solid tumors[2][7].
