This is a fixed combination therapy consisting of **zanidatamab**, a bispecific monoclonal antibody targeting two distinct epitopes on the extracellular domain of HER2, together with **fluorouracil**, **leucovorin**, and **oxaliplatin**, classic agents used in colorectal and gastrointestinal cancer chemotherapy protocols (notably "mFOLFOX6"). Zanidatamab exerts anti-tumor activity through HER2 signal blockade, induction of complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, phagocytosis, HER2 internalization, and downregulation. The regimen is under active clinical investigation primarily for HER2-positive advanced or metastatic gastroesophageal adenocarcinoma. The antibody portion (zanidatamab) is developed by Zymeworks and Jazz Pharmaceuticals. The combination is administered intravenously.
