This investigational combination therapy consists of zanidatamab, paclitaxel, and ramucirumab. Zanidatamab is a humanized bispecific monoclonal antibody that targets two non-overlapping epitopes (ECD2 and ECD4) of the Human Epidermal Growth Factor Receptor 2 (HER2), leading to dual HER2 signal blockade and enhanced antibody-dependent cellular cytotoxicity (ADCC). Ramucirumab is a fully human monoclonal antibody that targets Vascular Endothelial Growth Factor Receptor 2 (VEGFR2), acting as an anti-angiogenic agent by blocking the binding of VEGF ligands. Paclitaxel is a taxane-based chemotherapy agent that stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis. This triple combination is being developed by Jazz Pharmaceuticals and Zymeworks for the treatment of HER2-positive advanced gastroesophageal adenocarcinoma (GEA) in patients who have previously progressed on trastuzumab-containing regimens. It is currently being evaluated in the Phase 2 GA.4 clinical trial.
