Drug intelligence / Profile preview
Zanidatamab + tislelizumab is a combination of two monoclonal antibodies used experimentally in oncology. **Zanidatamab** is a humanized bispecific monoclonal antibody that targets two non-overlapping extracellular domains (domain 2 and domain 4) of HER2, leading to receptor clustering, internalization, downregulation, and induction of immune-mediated cytotoxicity, including antibody-dependent cellular cytotoxicity (ADCC), phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC). **Tislelizumab** is a humanized monoclonal antibody against PD-1; it blocks the PD-1/PD-L1 and PD-1/PD-L2 interactions, thereby reducing negative immune signaling and enhancing T-cell activity. The combination is under clinical investigation primarily for HER2-positive advanced or metastatic gastroesophageal adenocarcinoma, in both chemotherapy-containing and chemotherapy-free regimens[2][4][9].
