Zanidatamab zovodotin is a bispecific antibody-drug conjugate (ADC) targeting the human epidermal growth factor receptor 2 (HER2). It consists of a biparatopic IgG1 antibody (zanidatamab, also known as ZW25) that binds two distinct epitopes on HER2, conjugated to a cytotoxic microtubule inhibitor payload via proprietary linker technology. The drug is designed to deliver targeted cytotoxicity to HER2-expressing tumor cells by binding and internalizing into the cell, where the payload disrupts microtubule function leading to apoptosis. Zanidatamab zovodotin is being developed primarily for solid tumors with high or aberrant expression of HER2, including breast cancer, gastric cancer, and other solid tumors refractory to existing therapies. The developer and primary manufacturer is Zymeworks[1][3][8].
