Overview

zanolimumab + CHOP is a combination chemo-immunotherapy regimen investigated for the treatment of CD4-positive peripheral T-cell lymphoma (PTCL). It consists of zanolimumab (formerly HuMax-CD4), a humanized IgG1 monoclonal antibody targeting the CD4 surface antigen, and the standard CHOP chemotherapy backbone (cyclophosphamide, doxorubicin, vincristine, and prednisone). Zanolimumab works by binding to CD4-expressing malignant T cells, leading to their depletion via antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), while also potentially interfering with T-cell signaling. The CHOP components provide broad cytotoxic activity by damaging DNA and disrupting mitosis. Originally developed by Genmab and later acquired by Emergent BioSolutions via TenX BioPharma, the combination was evaluated in a Phase 2 trial (NCT00893516) for newly diagnosed nodal PTCL, though development was ultimately terminated.
