Overview

This is a multi-agent chemo-immunotherapy regimen combining the next-generation Bruton's tyrosine kinase (BTK) inhibitor zanubrutinib with the anti-CD20 monoclonal antibody rituximab and a backbone of high-dose methotrexate, liposomal doxorubicin, and etoposide. Developed by researchers at West China Hospital, Sichuan University, this intensive regimen is designed to improve outcomes in patients with primary central nervous system lymphoma (PCNSL) and secondary central nervous system lymphoma (SCNSL). The combination leverages the high CNS penetration of zanubrutinib and methotrexate alongside the cytotoxic effects of doxorubicin and etoposide to target B-cell malignancies within the central nervous system. It is currently being evaluated in Phase 2 clinical trials for its efficacy and safety in these difficult-to-treat lymphoma subtypes.
