Overview

This combination therapy regimen consists of zanubrutinib, a small-molecule Bruton tyrosine kinase (BTK) inhibitor, and CD19-directed chimeric antigen receptor (CAR) T-cell therapy. Zanubrutinib is administered as an adjunctive treatment to potentially enhance the efficacy, expansion, and persistence of the CAR T-cells. The CAR T-cell component involves autologous T cells genetically engineered to express a receptor targeting CD19, a protein expressed on the surface of B-cell lymphomas. The regimen is being investigated by Northwestern University for patients with recurrent or refractory aggressive B-cell non-Hodgkin lymphoma or transformed indolent B-cell lymphoma. The standard-of-care CAR T-cell products utilized in this combination include axicabtagene ciloleucel, tisagenlecleucel, or lisocabtagene maraleucel.
