Overview
**Zanzalintinib + pembrolizumab** is an investigational combination therapy for cancer treatment. Zanzalintinib is a novel, multi-targeted, small molecule tyrosine kinase inhibitor (TKI) that inhibits VEGFR (vascular endothelial growth factor receptor), MET, and the TAM kinases (TYRO3, AXL, MER), aiming to suppress tumor growth, angiogenesis, and promote an immune-permissive tumor microenvironment. Pembrolizumab (KEYTRUDA) is a monoclonal antibody immune checkpoint inhibitor that blocks PD-1 (programmed death-1) on T cells, enhancing the immune system's attack on cancer cells. This combination is under evaluation particularly for head and neck squamous cell carcinoma (HNSCC), with potential in other solid tumors. Early clinical data suggest the TKI-ICI regimen may offer improved efficacy and manageable safety compared to standard therapies[5][7].
