ZAP-70 siRNA is a small interfering RNA (siRNA) designed to silence the expression of Zeta-chain-associated protein kinase 70 (ZAP-70), a tyrosine kinase that is aberrantly expressed in aggressive forms of Chronic Lymphocytic Leukemia (CLL). In IGHV-unmutated CLL, ZAP-70 acts as a critical scaffold and signaling molecule that integrates tonic B cell receptor (BCR) signaling and chemokine-mediated pathways, such as those involving CCL19 and CCL21. By degrading ZAP-70 mRNA through the RNA interference (RNAi) pathway, this siRNA inhibits the constitutive activation of downstream kinases like AKT and GSK-3beta and impairs leukemia cell migration. While currently utilized primarily as a research tool in primary cell cultures and co-culture systems, it represents a targeted approach to disrupting the signaling networks that drive CLL progression.
