ZB014 is a half-life extended monoclonal antibody that dual-targets B-lymphocyte antigen CD19 and the inhibitory receptor FcγRIIb. Developed by Zenas BioPharma, the therapeutic is designed to treat autoimmune diseases by suppressing B-cell activation. By co-engaging CD19 and FcγRIIb, ZB014 leverages the natural inhibitory signaling of the FcγRIIb receptor to downregulate B-cell receptor-mediated signaling. The molecule features an engineered Fc domain to extend its circulating half-life, potentially allowing for less frequent dosing compared to first-generation antibodies. As of March 2026, ZB014 is in preclinical development.
