ZB021 is a novel, oral small molecule inhibitor targeting both the IL-17AA homodimer and the IL-17AF heterodimer. It is designed to block pro-inflammatory signaling mediated by IL-17AA and IL-17AF, which are implicated in a wide range of autoimmune diseases. ZB021 is intended for therapeutic use in rheumatology and dermatology, with strong preclinical data showing potent in vitro activity and robust in vivo efficacy. Its best-in-class potential is supported by favorable pharmacokinetic (PK) and absorption, distribution, metabolism, and excretion (ADME) properties, and preclinical efficacy comparable to reference anti-IL-17 biologics in animal models. The drug is advancing through IND-enabling studies, with a first-in-human study expected to begin in 2026. ZB021 is being developed by Zenas BioPharma, with in-licensed rights from InnoCare, and has worldwide rights outside Greater China and Southeast Asia.
