Overview

ZB131-DXd is an antibody-drug conjugate (ADC) consisting of ZB131, a humanized monoclonal antibody targeting cancer-specific plectin (CSP), conjugated to the topoisomerase I inhibitor deruxtecan (DXd). Developed by ZielBio, ZB131-DXd is designed to selectively target CSP, which is exclusively expressed on the surface of various cancer cells, including pancreatic, ovarian, and bile duct cancers, while remaining absent in normal or pre-malignant tissues. Upon binding to CSP, ZB131-DXd is rapidly internalized, leading to the intracellular release of the DXd payload, which induces DNA damage and subsequent apoptosis. Preclinical studies have demonstrated that ZB131-DXd has a drug-to-antibody ratio (DAR) of 7-8 and exhibits potent antitumor activity in xenograft models, supporting its potential clinical evaluation.
