ZC001

ZC001 is an antibody-drug conjugate (ADC) targeting Intercellular Adhesion Molecule 1 (ICAM-1/CD54), developed by Zhejiang Cancer Hospital for the treatment of advanced solid tumors, particularly esophageal squamous cell carcinoma (ESCC). The conjugate consists of a humanized anti-ICAM1 monoclonal antibody linked to the microtubule inhibitor monomethyl auristatin E (MMAE) via a cathepsin-cleavable valine-citrulline linker. ICAM-1 is a cell surface glycoprotein that is frequently overexpressed in ESCC and plays a role in tumor cell signaling and metastasis. ZC001 works by binding to ICAM-1, undergoing receptor-mediated endocytosis, and releasing MMAE within the lysosomal compartment, leading to mitotic arrest and apoptosis of the cancer cell.
