Overview

ZD2767P is a phenol mustard glutamate prodrug developed for use in Antibody Directed Enzyme Prodrug Therapy (ADEPT) for cancer. In this system, the prodrug ZD2767P is selectively activated at tumor sites by the bacterial enzyme carboxypeptidase G2 (CPG2), which can be targeted to tumors via antibody conjugation. Upon enzymatic cleavage by CPG2, the active cytotoxic agent ZD2767D (4-[N,N-bis(2-iodoethyl)amino]phenol), a bifunctional alkylating agent, is released and induces DNA double-strand breaks leading to apoptosis and ferroptosis in cancer cells. The approach aims to increase tumor specificity and reduce systemic toxicity compared to conventional chemotherapy. Recent studies have also explored ultrasound-enhanced delivery of this system (ZD2767P+CPG2+US), which further increases drug accumulation and efficacy against cisplatin-resistant cancers[1][3][5].
