ZD526 is an investigational small-molecule adenosine A2A receptor (A2AR) antagonist developed by Hangzhou Zhihe Dangu Biotechnology for the treatment of Alzheimer's disease (AD) and mild cognitive impairment (MCI). The adenosine A2A receptor is a G protein-coupled receptor that is highly expressed in the brain and plays a significant role in modulating neuroinflammation and synaptic plasticity. By selectively antagonizing this receptor, ZD526 aims to inhibit the overactivation of glial cells, reduce the release of pro-inflammatory cytokines, and protect neurons from amyloid-beta-induced toxicity, potentially slowing the progression of cognitive decline. Classified as a Class 1 innovative drug in China, ZD526 is administered orally and is currently undergoing clinical evaluation to assess its safety, pharmacokinetics, and preliminary efficacy in patients with cognitive disorders.
