Overview

ZD55 is a genetically modified oncolytic adenovirus (Ad5) characterized by the deletion of the E1B-55kD gene. This modification enables the virus to selectively replicate in and destroy cancer cells, particularly those with p53 pathway deficiencies, while sparing normal cells. Developed under the Cancer Targeting Gene-Virotherapy (CTGVT) strategy, ZD55 functions both as a direct oncolytic agent and a potent gene delivery vector. It is frequently engineered to carry various therapeutic antitumor genes (such as TRAIL, IL-24, or GM-CSF) to the tumor microenvironment, combining the strengths of viral oncolysis and gene therapy. Preclinical and early clinical studies have demonstrated its efficacy against a wide range of malignancies, including hepatocellular carcinoma, and its ability to target cancer stem-like cells.
