ZD6126 + oxaliplatin + 5-fluorouracil + leucovorin is a multi-agent combination chemotherapy regimen investigated primarily for cancer treatment. **ZD6126** is a vascular-targeting agent (a prodrug of N-acetylcolchinol) that disrupts tumor blood vessels and induces tumor necrosis. **Oxaliplatin** is a third-generation platinum compound that crosslinks DNA, causing inhibition of DNA replication and transcription, leading to cellular apoptosis. **5-fluorouracil** is an antimetabolite that inhibits thymidylate synthase, suppressing DNA synthesis and function. **Leucovorin** (folinic acid) is not cytotoxic alone but enhances the binding of 5-fluorouracil to thymidylate synthase, increasing its efficacy. This combination was studied for advanced cancers, notably colorectal cancer, to evaluate improvements in efficacy and tolerability and includes an investigational antiangiogenic mechanism with ZD6126 alongside DNA-damaging platinum and antimetabolite chemotherapies[4][2][1].
