Overview

ZD7288 is a small molecule pyrimidine derivative that serves as a potent and widely used pharmacological tool for blocking hyperpolarization-activated cyclic nucleotide-gated (HCN) channels. It inhibits the hyperpolarization-activated current (Ih), also known as the "funny" current (If) in the heart, which is critical for pacemaker activity and neuronal excitability. While ZD7288 is effective across all HCN subunits (HCN1-4), it is considered relatively non-specific and has been reported to exhibit off-target effects on voltage-gated sodium and calcium channels at higher concentrations. Originally developed by Zeneca (now AstraZeneca), the compound is primarily utilized in electrophysiological and developmental biology research, such as investigating the role of HCN channels in cardiac progenitor cell metabolism and ventricular chamber development.
