Drug intelligence / Profile preview
INCB161734 + SNV1521 is an investigational combination of two oral small-molecule targeted anticancer agents: zecdarasib (INCB161734), a selective non-covalent ON/OFF KRAS G12D inhibitor, and SNV1521, a selective, brain-penetrant PARP1 inhibitor. Developed through a clinical trial collaboration between Incyte and Synnovation, the combination is being evaluated in a Phase 1b clinical trial for the treatment of patients with advanced or metastatic pancreatic ductal adenocarcinoma (PDAC). The therapeutic rationale is to synergistically target KRAS G12D-driven oncogenic signaling while simultaneously inhibiting PARP1-mediated DNA damage repair, thereby enhancing genomic instability and tumor cell apoptosis in KRAS G12D-mutated pancreatic cancers.
