Zenagamtide (formerly known as amycretin) is a long-acting, unimolecular co-agonist of the glucagon-like peptide-1 (GLP-1) receptor and the amylin receptor, developed by Novo Nordisk for the treatment of obesity and overweight. By simultaneously activating both the GLP-1 and amylin pathways, zenagamtide aims to provide enhanced weight loss efficacy compared to single-receptor agonists. GLP-1 receptor activation primarily reduces appetite and slows gastric emptying, while amylin receptor activation promotes satiety and regulates glucose metabolism. The drug is currently being evaluated in Phase 3 clinical trials (the AMAZE program) as a once-weekly subcutaneous injection, following promising Phase 1 results that demonstrated significant weight reduction in participants with obesity.
