Overview

Zeniplatin is a third-generation, water-soluble platinum complex developed as an antineoplastic agent for the treatment of refractory and advanced solid malignancies, including ovarian cancer, malignant melanoma, and renal cell cancer[1][2][3][4][5]. Its mechanism of action is similar to other platinum-based chemotherapy agents: zeniplatin acts as a **DNA alkylating agent**, forming adducts with DNA, which inhibits DNA replication and transcription, ultimately leading to apoptosis in rapidly dividing tumor cells[3]. Its dose-limiting toxicity is primarily hematological (mainly leukopenia and neutropenia), along with notable non-hematological side effects such as nausea and vomiting. Zeniplatin was evaluated in multiple phase I and II clinical trials, demonstrating some efficacy—particularly in malignant melanoma and ovarian cancer—but development was discontinued due to unacceptable nephrotoxicity observed in later trials, even with prophylactic hydration[2][3][4].
