Overview

Zenocutuzumab + enzalutamide is an investigational combination therapy comprising **zenocutuzumab**, a bispecific monoclonal antibody that targets the HER2 and HER3 receptors and inhibits neuregulin 1 (NRG1) fusion oncogenic signaling, and **enzalutamide**, a next-generation androgen receptor (AR) signaling inhibitor. Zenocutuzumab blocks NRG1-driven tumor growth by preventing HER2/HER3 dimerization and signaling, while also recruiting immune effector cells to destroy tumor cells. Enzalutamide antagonizes the androgen receptor, inhibiting androgen-dependent cell proliferation. This combination is under clinical investigation for **metastatic castration-resistant prostate cancer (mCRPC)** to potentially overcome resistance and enhance antitumor effects, especially in patients who have progressed on AR path inhibitors[1][3][5].
