This combination therapy consists of three antiretroviral agents: stavudine (Zerit), lamivudine (Epivir), and nevirapine (Viramune). Stavudine and lamivudine are nucleoside reverse transcriptase inhibitors (NRTIs) that act as chain terminators during viral DNA synthesis by competing with natural substrates for incorporation into the viral DNA strand. Nevirapine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that binds directly and non-competitively to the HIV-1 reverse transcriptase enzyme, inducing a conformational change that inhibits its catalytic activity. This specific regimen was utilized as the innovator reference arm in a bioequivalence study conducted by Makerere University in Uganda to compare the branded individual components against the generic fixed-dose combination Triomune in HIV-infected adults.
