Overview

Zeta-MBC-005 (also known as ZetaMAST) is a proprietary injectable drug-eluting hydrogel carrier developed by Zetagen Therapeutics for the locoregional treatment of unresectable, multifocal breast cancer metastases to the liver. The hydrogel is designed for controlled release of small molecule tumoricidal agents directly at metastatic sites in the liver. Preclinical studies have demonstrated that two concentrations of Zeta-MBC-005 significantly reduced tumor burden and increased survival rates compared to doxorubicin in mouse models with triple-negative breast cancer (TNBC) liver metastases. The therapy aims to address an unmet need in patients with metastatic breast cancer involving the liver—a population with poor prognosis and limited curative options—by providing local control and potential systemic biological effects. As of June 2025, it remains in preclinical development with plans for a Phase 1b clinical trial.
