Overview

Zevaquenabant (INV-101) is a first-in-class, peripherally acting small molecule inverse agonist/antagonist of the cannabinoid receptor 1 (CB1). It is being developed primarily for the treatment of idiopathic pulmonary fibrosis (IPF), including progressive fibrotic interstitial lung diseases such as systemic sclerosis and Hermansky-Pudlak Syndrome. The drug acts by blocking CB1 receptors outside the central nervous system, aiming to reduce fibrosis while minimizing CNS-related side effects seen with earlier CB1 blockers. Zevaquenabant has also been investigated for Prader-Willi syndrome (PWS) and non-alcoholic steatohepatitis (NASH), and has received FDA rare pediatric disease designation for PWS. Preclinical studies have shown that it reduces fibrosis in animal models comparably to nintedanib, a current standard therapy for IPF[1][2][3][5][7].
