Zevotrelvir is an orally administered small-molecule inhibitor of the SARS-CoV-2 main protease (Mpro, also known as 3CL protease), developed by Enanta Pharmaceuticals as a direct-acting antiviral for the treatment of COVID-19. It is a non-covalent, nanomolar 3CLpro inhibitor with a spirocyclic indole-based scaffold that selectively targets the viral protease essential for processing the SARS-CoV-2 polyproteins, thereby blocking viral replication with broad activity against human coronaviruses in biochemical assays.[1][4][5][7][9][11][13][15]
