Overview

ZFPG is an engineered nanozyme nanoparticle consisting of a ZnFe2O4@Pt core and a glucose oxidase (GOx) shell, developed for the treatment of prostate carcinoma. It utilizes a multi-modal therapeutic strategy combining cascade-enzyodynamic therapy, sonodynamic therapy, and metallo-immunotherapy. The nanoparticles exhibit five-enzyme activities and sonosensitivity, allowing them to deplete glutathione (GSH) and glucose within the tumor microenvironment while generating multiple bursts of reactive oxygen species (ROS) upon ultrasound stimulation. This oxidative stress upregulates heme oxygenase 1 (HMOX1), leading to the accumulation of ferrous ions (Fe2+) and lipid peroxides, which triggers immunogenic ferroptosis. Additionally, ZFPG NPs possess magnetic targeting properties to enhance tumor accumulation and can remodel the immunosuppressive tumor microenvironment to stimulate anti-tumor immunity and inhibit metastatic progression.
