Zfra1-31 is a synthetic 31-amino-acid peptide corresponding to the full-length Zinc finger-like protein for regulating apoptosis (Zfra). It functions as a regulator of apoptosis by participating in the mitochondrial pathway and modulating the activity of tumor suppressors such as WWOX and p53. Zfra1-31 induces a novel form of protein degradation termed "zfration," which occurs independently of the ubiquitin-proteasome system. It has been shown to suppress the phosphorylation of WWOX at Ser14, thereby inhibiting the docking and growth of metastatic melanoma cells in the lung and liver. Additionally, it interacts with membrane hyaluronidase Hyal-2 and stimulates specific memory lymphocytes. Preclinical studies in mouse models have demonstrated its potential in preventing skin cancers, including melanoma and basal cell carcinoma.
